Antibiotics produced by Streptomyces BJID
Procópio,Rudi Emerson de Lima; Silva,Ingrid Reis da; Martins,Mayra Kassawara; Azevedo,João Lúcio de; Araújo,Janete Magali de.
Streptomyces is a genus of Gram-positive bacteria that grows in various environments, and its shape resembles filamentous fungi. The morphological differentiation of Streptomyces involves the formation of a layer of hyphae that can differentiate into a chain of spores. The most interesting property of Streptomyces is the ability to produce bioactive secondary metabolites, such as antifungals, antivirals, antitumorals, anti-hypertensives, immunosuppressants, and especially antibiotics. The production of most antibiotics is species specific, and these secondary metabolites are important for Streptomyces species in order to compete with other microorganisms that come in contact, even within the same genre. Despite the success of the discovery of antibiotics,...

Antimicrobial activity of actinomycetes and characterization of actinomycin-producing strain KRG-1 isolated from Karoo, South Africa BJPS
Charousová,Ivana; Medo,Juraj; Hleba,Lukáš; Císarová,Miroslava; Javoreková,Soňa.
In the present study we reported the antimicrobial activity of actinomycetes isolated from aridic soil sample collected in Karoo, South Africa. Eighty-six actinomycete strains were isolated and purified, out of them thirty-four morphologically different strains were tested for antimicrobial activity. Among 35 isolates, 10 (28.57%) showed both antibacterial and antifungal activity. The ethyl acetate extract of strain KRG-1 showed the strongest antimicrobial activity and therefore was selected for further investigation. The almost complete nucleotide sequence of the 16S rRNA gene as well as distinctive matrix-assisted laser desorption/ionization-time-of-flight/mass spectrometry (MALDI-TOF/MS) profile of whole-cell proteins acquired for strain KRG-1 led to...

Biodiversity of Antimicrobial-Producing Actinomycetes Strains Isolated from Dry Dipterocarp Forest Soil in Northeast Thailand BABT
Chanthasena,Panjamaphon; Nantapong,Nawarat.
The increasing use of antibiotics has led to the development of drug-resistant microorganisms. The emergence of resistant strains has compromised the treatment and control of infectious diseases. As a result, the search and development of a novel class of antibiotic drugs are required. Actinomycetes have been recognized as a richest source of secondary metabolites including antibiotics. The aim of this study was to investigate the diversity of antibiotic-producing soil Actinomycetes from dry dipterocarp forest in Northeast of Thailand. The soil from this area has been known for its poor in nutrients and highly acidic with pH values around 4.5. In such a harsh condition, soil-inhabiting Actinomycetes elevate their protective mechanisms by inducing the...

Biological control of phytopathogenic fungi by endophytic actinomycetes isolated from maize (Zea mays L.) BABT
Costa,Francisco Gheler; Zucchi,Tiago Domingues; Melo,Itamar Soares de.
This work aimed a survey on the biodiversity of maize endophytic actinomycete, and an evaluation of their potential to control the phytopathogenic fungi. From several regions of São Paulo state, 40 strains were isolated from the healthy maize plants. The identification of these strains, based on morphological properties and fatty acid methyl ester (FAME) profile showed that most of them belonged to the Streptomyces genus. These isolates were first screened for the growth inhibition of phytopathogenic fungi and results showed that all the isolate were able to inhibit the development of at least one tested pathogen. Two selected isolates were then evaluated for the control of P. aphanidermatum in cucumber (Cucumis sativa L.) under greenhouse conditions....

Carbon catabolite repression of retamycin production by Streptomyces olindensis ICB20 BJM
Inoue,Olavo Ossamu; Schmidell Netto,Willibaldo; Padilla,Gabriel; Facciotti,Maria Cândida Reginato.
Retamycin is an anthracyclinic antitumoral complex produced by Streptomyces olindensis ICB20. In this work the influence of different glucose concentrations in the feed medium on the production of retamycin was studied. Chemostat cultures employing glucose concentration varying between 10 g/L and 25 g/L showed that use of high glucose concentration resulted in catabolite repression of the biosynthesis of the antitumoral. The highest specific retamycin production rate, qRTM = 7.8 mg/g.h, was obtained when glucose concentration was 10 g/L. The lowest value of qRTM, 2.5 mg/g.h, was observed when glucose concentration was 20 and 25 g/L. The residual glucose concentration varied from 0 to 13 g/L, as the glucose concentration in the feed was increased from 10 to...

ABSTRACT Here, we show the draft genome sequence of Streptomyces sp. F1, a strain isolated from soil with great potential for secretion of hydrolytic enzymes used to deconstruct cellulosic biomass. The draft genome assembly of Streptomyces sp. strain F1 has 69 contigs with a total genome size of 8,142,296 bp and G + C 72.65%. Preliminary genome analysis identified 175 proteins as Carbohydrate-Active Enzymes, being 85 glycoside hydrolases organized in 33 distinct families. This draft genome information provides new insights on the key genes encoding hydrolytic enzymes involved in biomass deconstruction employed by soil bacteria.

Effects of Brassica napus seed meal amendment on soil populations of resident bacteria and Naegleria americana, and the unsuitability of arachidonic acid as a protozoan-specific marker OAK
Cohen, Michael F.; Mazzola, Mark.
Bacterial and protozoan populations were monitored in orchard soil amended with Brassica napus seed meal. Numbers of fluorescent pseudomonads peaked one week following incorporation of the amendment and declined thereafter, whereas populations of Streptomyces spp. increased to stable levels nearly 20-fold higher than in non-treated soil. The amoeba-flagellate Naegleria americana, isolated from the amended soil, showed a marked feeding preference in culture for fluorescent pseudomonads relative to Streptomyces spp. Amended soils showed rapid declines in RSM-derived cis-oleic acid that paralleled increases in the numbers of bacteria and proportion of bacterial fatty acid markers in the soil. Arachidonic acid, which reached peak levels one week after seed...

Ser,Hooi-Leng; Tan,Wen-Si; Ab Mutalib,Nurul-Syakima; Yin,Wai-Fong; Chan,Kok-Gan; Goh,Bey-Hing; Lee,Learn-Han.
ABSTRACT As the largest genus in Actinobacteria family, Streptomyces species have the ability to synthesize numerous compounds of diverse structures with bioactivities. Streptomyces mangrovisoli MUSC 149T was previously isolated as a novel streptomycete from mangrove forest in east coast of Peninsular Malaysia. The high quality draft genome of MUSC 149T comprises 9,165,825 bp with G + C content of 72.5%. Through bioinformatics analysis, 21 gene clusters identified in the genome were associated with the production of bioactive secondary metabolites. The presence of these biosynthetic gene clusters in MUSC 149T suggests the potential exploitation of the strain for production of medically important compounds.

Glucose(xylose) isomerase production by Streptomyces sp. CH7 grown on agricultural residues BJM
Chanitnun,Kankiya; Pinphanichakarn,Pairoh.
Streptomyces sp. CH7 was found to efficiently produce glucose(xylose) isomerase when grown on either xylan or agricultural residues. This strain produced a glucose(xylose) isomerase activity of roughly 1.8 U/mg of protein when it was grown in medium containing 1% xylose as a carbon source. Maximal enzymatic activities of about 5 and 3 U/mg were obtained when 1% xylan and 2.5% corn husks were used, respectively. The enzyme was purified from a mycelial extract to 16-fold purity with only two consecutive column chromatography steps using Macro-prep DEAE and Sephacryl-300, respectively. The approximate molecular weight of the purified enzyme is 170 kDa, and it has four identical subunits of 43.6 kDa as estimated by SDS-PAGE. Its Km values for glucose and...

Hata,Erneeza Mohd; Sijam,Kamaruzaman; Ahmad,Zainal Abidin Mior; Yusof,Mohd Termizi; Azman,Noor Aisyah.
ABSTRACT The aim of this work was to invitro assay the antimicrobial activity of actinomycetes in rice against Xanthomonas oryzae pv. oryzicola and as potential plant growth promoter. A total of 92 actinomycete strains were isolated from different rice plant components and field locations. Of these, only 21.74% showed antagonistic activity against the Xoc pathogen. Molecular identification via 16s rRNA amplification revealed that 60% of the active antagonistic strains belonged to the genus Streptomyces. Isolates that demonstrated the highest antagonistic activity were also able to produce hydrolytic enzymes and plant growth-promoting hormones. Combination of preliminary screening based on in vitro antagonistic, hydrolytic enzyme and plant growth hormone...

Poly R decolorization and APPL production by Streptomyces violaceoruber and Streptomyces spiroverticillatus BJM
Abou-Dobara,M.I.; Omar,N.F..
Two mesophilic streptomycetes (S. violaceoruber and S. spiroverticillatus) were selected to study their Poly R-478 decolorization ability and lignocellulose solubilizing activity. Both strains were able to degrade Poly R-478 dye and ferulic acid during growth on a minimal salts medium. The Poly R-478 decolorizing activities of both strains were induced by adding different carbon sources to the culture media. S. violaceoruber could decolorize 63% of Poly R-478 after 24 h. Both strains could solubilize straw and produce acid-precipitable polymeric lignin (APPL) with different efficiency. From the major extracellular enzymes recovery of both strains on rice and wheat straw, we can predicate that the biodegradation process was partial indicating a possible...

Production and characterization of a biosurfactant produced by Streptomyces sp. DPUA 1559 isolated from lichens of the Amazon region BJMBR
Santos,A.P.P.; Silva,M.D.S.; Costa,E.V.L.; Rufino,R.D.; Santos,V.A.; Ramos,C.S.; Sarubbo,L.A.; Porto,A.L.F..
Surfactants are amphipathic compounds containing both hydrophilic and hydrophobic groups, capable to lower the surface or interfacial tension. Considering the advantages of the use of biosurfactants produced by microorganisms, the aim of this paper was to develop and characterize a biosurfactant produced by Streptomyces sp. DPUA1559 isolated from lichens of the Amazon region. The microorganism was cultured in a mineral medium containing 1% residual frying soybean oil as the carbon source. The kinetics of biosurfactant production was accompanied by reducing the surface tension of the culture medium from 60 to values around 27.14 mN/m, and by the emulsification index, which showed the efficiency of the biosurfactant as an emulsifier of hydrophobic compounds....

Streptomyces globosus DK15 and Streptomyces ederensis ST13 as new producers of factumycin and tetrangomycin antibiotics BJM
Charousová,Ivana; Medo,Juraj; Hleba,Lukáš; Javoreková,Soňa.
ABSTRACT Fifty seven soil-borne actinomycete strains were assessed for the antibiotic production. Two of the most active isolates, designed as Streptomyces ST-13 and DK-15 exhibited a broad range of antimicrobial activity and therefore they were selected for HPLC fractionation against the most suppressed bacteria Staphylococcus aureus (ST-13) and Chromobacterium violaceum (DK-15). LC/MS analysis of extracts showed the presence of polyketides factumycin (DK15) and tetrangomycin (ST13). The taxonomic position of the antibiotic-producing actinomycetes was determined using a polyphasic approach. Phenotypic characterization and 16S rRNA gene sequence analysis of the isolates matched those described for members of the genus Streptomyces. DK-15 strain exhibited...

Streptomyces lunalinharesii 235 prevents the formation of a sulfate-reducing bacterial biofilm BJM
Rosa,Juliana Pacheco da; Tibúrcio,Samyra Raquel Gonçalves; Marques,Joana Montezano; Seldin,Lucy; Coelho,Rosalie Reed Rodrigues.
ABSTRACT Streptomyces lunalinharesii strain 235 produces an antimicrobial substance that is active against sulfate reducing bacteria, the major bacterial group responsible for biofilm formation and biocorrosion in petroleum reservoirs. The use of this antimicrobial substance for sulfate reducing bacteria control is therefore a promising alternative to chemical biocides. In this study the antimicrobial substance did not interfere with the biofilm stability, but the sulfate reducing bacteria biofilm formation was six-fold smaller in carbon steel coupons treated with the antimicrobial substance when compared to the untreated control. A reduction in the most probable number counts of planktonic cells of sulfate reducing bacteria was observed after treatments...
